Instead of drilling a hole on the bottom of my acrylic tank and the stand (A), would drilling the back and going around my stand work? (B)

yup - will work just fine - in fact any tank with a tempered glass bottom has to have the holes in the back! it just adds a couple more pieces/parts to the plumbing

I would just drill the back like B but where you want the water line, put a strainer in the overflow, and put the durso externally to the tank.

That is how I did mine. Works fine.

That will work great, plus if your overflow fails, bulkhead fails etc, you will not drain the hole tank.

45 degree elbows will allow for more flow but not really necessary since it is only a drain.
