happy holidays
I have a 210 gallon tank with sensitive fish (no coral) and barebottom.
I been getting ammonia spike and plan to add sand back into the tank for a biological filter.
The question is should I add slowly a few pounds a day or put it all at once?

If you plan on a DSB using LS then add about a half inch every other day until you reach the desired be height. It lets some of the top dwelling critters some time to climb up without being crushed. For base sand you can add it all at once and place the LS on top.

oh yes, what would be the desirable height for a biological filter?

I usually suggest at least 4".

Why are you getting ammonia spikes? I would figure that out before adding sand. There are plenty of reef keepers with bare bootom tanks that never have amonia spikes. How much live rock do you have? Is your tank overstocked with fish and corals? Is your filtration equipment working properly and is it big enough for your tank? Just some things to think about. There are many reef hobbyist with bare bootom tanks that never have ammonia spikes.
