I just don't have the patience to shake the test tube after each drop when using my API test kit. Is it okay to load up on the first 15-20 drops or so to save time testing calcium?

Yes. I believe it says that if you think you calc is around 400 to add 15 drops all at once.

+1 I've tried it both ways and always got the same results.

Ditching the test vials that come with the API kits help as well... I use Salifert vials with a 5ml syringe so I can swirl and mix each drop easily.

I've gotten some very strange results when I start by adding 10 drops or so before going one at a time. Same thing with Alkalinity tests. I found I got false high readings by doing that.
I now recap and shake between each drop, but that's just me.
