I have a pre-drilled 75 gallon that has the drain on the back left corner and the return on the back right corner. For the return, could I plug the existing hole and bring the return over the rim of the tank? I played around with the idea of building a box to a hide a return pipe in that corner, but it will take up too much room. Someone suggested a pipe and return in tank, but I don't like that look. Any idea if over the top would work?

Yes, it would work fine. You can plug the hole with a piece of glass siliconed over the hole.

Use a Bulkhead fitting with female threads and thread a plug into it. Your plans for that return hole may change in the future, so why permanently seal it? You can come over the top with your return line. You can tee it and have it return to both sides of the tank or connect it to a squid(carefull with the sqwd, slows flow).

Instead of plugging it, why not use the return as an emergency/backup drain?
