Yes, really wouldn't be any different than doing a water change in my opinion.... I'm about to add one to my 34g AIO that's been going for about 4 months.... Did the same on my 55 after it had been up for a while, with out no ill effects...

I added a sump to my existing tank and wish I had done it sooner. The hardest part is the overlow. You will need an overflow that goes over the rim if the tank is up and running, unless you are brave enough to try to drill a full tank (I wasn't).
