I'd like to get a bigger sump than what I have now. Should I replace the live sand completely or put new sand in and transfer the old. Would I be able to use the mud instead?

If you are happy with the old sand use it. I would just have some new Saltwater ready and after the sand is moved try to siphon as much of the cloudy water out of the sump and replace it with the new water.

so it won't effect the biological filtration at all by putting new substrate in there?

I would put new sand in & transfer the old, your not affecting the biological filtration in a negative way, if anything you will be increasing the area that the beneficial de-nitrifying bacteria has to colonize.....what I would do is go ahead & add the new sand 1st, then add the existing sand, fill up your sump but allow it to settle for a bit before hooking it back up to your main tank.....this will reduce the amount of debris/dust that will be released into your main tank.....
