I am going to be doing the plumbing on my new tank, 45 X 24 X 20. What are the advantages / disadvantages of plumbing the drain / return using PVC rather than flexible tubing? Any feedback experience would be appreciated thanks....

Spaflex uses the regular PVC slip-fit fittings, and doesn't kink

Tubing helps reduce vibration from the pump, which makes it quieter. Tubing is also easier on head loss compared to using 90 or 45 degree bends with PVC.

I like the tubing for everything Mike321 said plus there's only 2 places for it to leak, at each ends connection (easy fix). I like that it's clear and I can spot occlusions. plus it's easy. :)
