Sumpless for 48 hours?

I have to redo my sump, is it safe to shut off the ball valves to the sump and repair the sump? And just have my Vortech MP40's run in the tank for waterflow. Just want to make sure before I tackle it.

I am using GE 100% silicone, it says 12 hours to cure. Should I wait 24-48 hours before re-filling the sump for operation?
