I currently have 1 drain and one return coming through my corner overflow. I want to make the return another drain and add a hob return. When I glue my PVC pieces, do I have to wait for them to dry and cure before running water through the pipes or can I attach my pieces and run my plumbing? This is a running tank and sump.

I would let the dry and cure as per the directions before putting them online.

There are different types of PVC glue. Look for the fast drying type to minimize downtime.

I have used PVC within minutes of connecting, though not ideal I suffered no ill effects.
Here is a table of cure times:

We don't use high pressure in the hobby usually(especially drains)....so a couple minutes is fine for handling and 1 hour sufficient for running it.
