How much of a budget do you have. Water/mold resistant sheet rock is a step above, a good primer with gloss finish will work, if you have mold issues it's an air flow problem not so much a drywall issue.

There are many options to choose from but you'll be limited if the wall is supporting weight and what your budget is. I really like the full view from both sides but cleaning is terrible if you don't have access on both sides. I like the porthole concept but not sure how you would clean them and not sure how a 4" gap between the hole and the tank will turn out.
The wall is not a supporting wall.

Budget is always a concern, so the less the better, but I'd rather do it right over time than poorly over night.

What I would do is cut the 2x4's out completely, and have the tank butted up against the wall with the port holes. But the glass cleaning process is an issue.
